چکیده
Direct immunofluorescence and direct immunoperoxidase staining were equally sensitive and specific for detection of herpes simplex virus antigen in lesion specimens, and each method showed 82% agreement with virus isolation results.

Comparison of direct immunoperoxidase and direct immunofluorescence for the detection of herpes simplex virus antigen in cell culture.
150 specimens from suspected herpes simplex genital and skin lesions were received in virus transport medium. They were inoculated into Hep-2 cell monolayers, examined for the presence of virus by cytopathic effect (CPE), direct immunoperoxidase (DIP) and direct immunofluorescence (DIF). Of 39 (26.0%) virus-positive specimens by CPE, 37 (24.7%) were HSV-positive by DIP and 36 (24.0%) by DIF sta...
متن کاملDirect immunofluorescence staining for detection of herpes simplex and varicella-zoster virus antigens in vesicular lesions and certain tissue specimens.
Direct immunofluorescence (IF) staining was compared with virus isolation for detection of herpes simplex viruses (HSV) and varicella-zoster virus (VZV) directly in clinical materials. These included 199 vesicular lesion specimens and 280 tissue specimens. Correspondence between IF and isolation results was 88% in testing for HSV in lesion specimens and 98% in testing for HSV in various tissue ...
متن کاملImmunofluorescence staining for the diagnosis of herpes encephalitis.
Direct immunofluorescence staining for herpes simplex virus was applied to cryostat sections of 43 specimens removed at brain biopsy. Herpes antigen was found in 10 specimens and virus was isolated from them. Antigen was found in one specimen from which virus was not isolated. Two specimens from which virus was isolated gave equivocal fluorescence. Thirty specimens gave no fluorescence or live ...
متن کاملLocalization of Herpes Simplex Virus Type 1 DNA in Latently Infected BALB/c Mice Neurons Using in situ Polymerase Chain Reaction
Background: Herpes simplex virus type-1 (HSV-1) establishes a lifelong latent infection in neurons following primary infection. The existence of latent HSV-1 DNA in the trigeminal ganglia of infected BALB/c mice was examined using a direct in situ PCR technique, based on Digoxigenin-11-dUTP detection system with anti-digoxigenin-peroxidase and 3,3'-diaminobenzidine (DAB) substrate. Methods: Eig...
متن کاملComparison of in situ hybridization and immunologic staining with cytopathology for detection and identification of herpes simplex virus infection in cultured cells.
Two recently developed sensitive techniques, in situ hybridization with a biotinylated cloned DNA probe and an avidin-biotin complex immunoperoxidase assay, were compared with the appearance of cytopathic changes for the early detection of herpes simplex virus infection in cell culture. By using commercially made reagents, these detection methods were evaluated in two different cell culture sys...
